In reply, Rupganj lost opener Azmir Ahmed in the second over but number three batsman Mominul Haque scored 55 and kept the scoreboard ticking before getting out in the 18th over.
Experienced batsman Shahriar Nafees (25) also got out in the 20th over and Rupganj were in trouble.
But captain Naeem Islam and Indian recruit Rishi Dhawan formed a 99-run partnership for the fifth wicket to get them back on track.
Dhawan scored 51 before getting out. Then Nayeem formed another crucial partnership of 71 runs off just 57 balls with wicketkeeper batsman Jaker Ali.
Naeem scored 85 off 92 balls with 8 boundaries before losing his wicket in the 48th over.
Eventually, Rupganj reached the target with four balls to spare Jaker Ali remain unbeaten on 34 off 26 balls.
Skipper Naeem was adjudged man-of-the-match for his match-winning knock.
